With the advancement of science, the research of pharmaceutical preparations has entered the era of drug release systems. Future research on drug delivery systems will play a decisive role in the fields of chemical and pharmaceutical, biotechnology or biotherapy. The lag in the development of pharmaceutical excipients has become an important factor restricting the development of China's drug delivery system. Some experts suggested that the role of pharmaceutical excipients in formulation research should be emphasized, and support should be increased to promote the rapid and healthy development of the domestic excipient industry.
Since the drug delivery function of the drug delivery system is entirely supported by excipients, the research and application of new excipients, the re-evaluation of existing excipient functions and the research of new formulation processes should become the first-selected scientific research projects of pharmaceutical companies in China. Researcher Shen Huifeng, a senior pharmaceutical preparation expert in China, believes that the active drug in the pharmaceutical dosage form is the substantial main part and determines the entire direction of action; the excipients ensure that the drug is selectively delivered to the tissue site in a certain procedure to prevent the drug from being lost before being released from the main body Live, and let the drug be released in a certain location in the body at a certain speed and time. Therefore, the dosage form composed of appropriate excipients has a positive and key role in the practical application of drugs and the effect of therapeutic effects.
In recent years, the role of medicinal excipients in the drug delivery system has been increasingly strengthened and is widely used. For example, the design and manufacture of oral sustained / controlled release drug delivery systems mainly use polymer materials, such as ethyl cellulose, light propyl methyl cellulose, polyacrylic acid resin vinegar series, etc., and chitosan and glycoproteins, etc. are also used. The application of controlled-release inclusion compounds is also increasing. As another example, microcapsules and microparticle-type controlled-release preparations often use polymer materials such as gelatin, starch, albumin, polylactic acid, polyglycolic acid-lactic acid copolymer, polymethylphthalamide, polypropylene-based acetone, ethyl cellulose, etc. ; Biodegradable polymers (copolymers of polylactic acid, polyglycolic acid and polyglycolic acid monolactic acid) applied to the delivery of protein and skin drugs can reduce the dosage of administration, increase the stability of the protein and achieve localization and constant speed release ; Both dextran and chitosan can be decomposed under the action of bacterial enzymes in the colon, and are not easily hydrolyzed in the stomach and small intestine. It is a suitable material for the colon targeted drug delivery system; used for the colon targeted adhesive drug delivery system Most of the carriers are non-specific carriers, such as alginate, cellulose derivatives, dextran, gelatin, pectin, chitin, etc. Among them, carbomer is the most widely used; methacrylic acid copolymer, light propyl methyl Cellulose acetate boric acid, cellulose acetate phthalate, etc. can be used as enteric materials; cyclodextrin is decomposed into oligosaccharides by the fermentation of colonic microorganisms, but it is not easily hydrolyzed, so it will not be in the stomach And the drug released in the small intestine can be used to prepare a colon targeted drug delivery system.
"China's pharmaceutical industry has been developing heavy raw materials and researching light preparations for a long time. China's drug release technology still has a large gap compared with the world's advanced level. Taking slow and controlled release preparations as an example, there are more than 200 foreign countries. There are less than 100 kinds of normal production in China. "Tu Jiasheng, a professor at the School of Pharmacy, China Pharmaceutical University, pointed out that the research on drug release systems in China is mainly traceable, and there are not many development and application of new drug release technologies. There are many reasons for the lag in development, but the reasons for the auxiliary materials cannot be ignored. In terms of the development of traditional Chinese medicine delivery systems, in addition to the complex composition of traditional Chinese medicine preparations, the material basis of the efficacy is difficult to determine; the traditional Chinese medicine preparation process is relatively backward, the scientific and technological content is not high, the competitiveness is weak, and the added value is low; research on compound preparations still exists In addition to some difficulties and other factors, new excipients have not yet been developed and applied to traditional Chinese medicine preparations are also important influencing factors. At present, although more than a dozen new pharmaceutical excipients have been used in traditional Chinese medicine preparations, the only ones that are actually used are cyclodextrin and sodium methylcellulose.
Tu Jiasheng believes that there are quite a few domestic excipients whose quality is unstable and can not meet the requirements of drug release technology. Therefore, a part of the new sustained-release new dosage form drugs can only stay in the laboratory, cannot be mass-produced, and it is difficult to enter the market. In addition, China's research and development of new excipients and quality control of pharmaceutical excipients also lag far behind developed countries. He suggested that the relevant state departments should actively organize, research units and enterprises should work closely together, scientific planning, unified management, and strengthen research on the quality of pharmaceutical excipients. We should focus on the development of related auxiliary materials for various formulations, such as new binders, disintegrants, coating materials, pigments, co-solvents, surfactants, etc. Strengthen the research and development of enteric-coated excipients of cellulose derivatives, soybean lecithin for injection, incubating phospholipid, and poloxamer for injection. In addition, through policy encouragement and other means, accelerate the rapid and healthy development of China's auxiliary materials industry.
